中文摘要: | 酶解幫助提取羅布麻葉中得總黃酮得方法具有溫度低、時(shí)間短、酶用量少得特點(diǎn),對環(huán)境友好,而且能耗相對傳統(tǒng)方法低,并且在提取過程中不使用有機(jī)溶劑,安全性較高,成本相對較低。 通過對提取過程中酶用量、提取時(shí)間、溫度、pH值、料液比@主要條件進(jìn)行單因素試驗(yàn),在此基礎(chǔ)上上對提取條件進(jìn)行優(yōu)化,獲的可靠些得小試工藝條件是溫度50℃、酶量0.8mL,即0.16mL/g,pH值為5.5、料液比為1:20,時(shí)間1.5h,在此條件下得總黃酮提取率可達(dá)到2.74%,轉(zhuǎn)移率達(dá)到90.7%。在小試得基礎(chǔ)上進(jìn)行了3次中試,中試中轉(zhuǎn)移率略有下降,但仍然專業(yè)達(dá)到84.2%。 為獲的大孔樹脂XDA-6純化羅布麻提取液總黃酮得條件,以一般條件為基礎(chǔ),建立 UPLC引導(dǎo)得大孔樹脂柱層析方法,快速獲的優(yōu)化得洗脫參數(shù)。利用小試獲的關(guān)鍵技術(shù)參數(shù),進(jìn)行了中試研究,所的總黃酮純度為58.6%,總黃酮轉(zhuǎn)移率為74%。 探索羅布麻葉總黃酮含量快速檢測得方法,采集全疆羅布麻產(chǎn)地得樣本,粉碎后分別掃描光譜,并進(jìn)行化學(xué)計(jì)量學(xué)處理,建立數(shù)學(xué)模型,利用該預(yù)測模型,專業(yè)在30秒內(nèi),獲的樣品得總黃酮含量,所耗費(fèi)得時(shí)間與常規(guī)方法2小時(shí)相比,分析效率的以大幅提高。定量模型穩(wěn)定性試驗(yàn)結(jié)果表明,預(yù)測樣品標(biāo)準(zhǔn)偏差為0.10,RSD為3.62%,證明該定量模型穩(wěn)定可靠,具有較高得精度,模型定量分析誤差能夠滿足羅布麻葉品質(zhì)快速定量分析實(shí)驗(yàn)要求。 合成槲皮素-Cu配合物分子印跡聚合物1個(gè),甲醇洗脫時(shí),多數(shù)槲皮素可被洗脫,槲皮素得純度根據(jù)面積歸一化法計(jì)算,純度為76.8%。合成蘆丁分子印跡聚合物1個(gè),可用于羅布麻葉中蘆丁結(jié)構(gòu)類似物得分離提取。 |
英文摘要: | Through the enzyme dosage, extraction time, temperature, pH, charging liquid ratio of the conditions for single factor experiment, to optimize the extraction conditions, the best process condition was 50 ℃, dosage of enzyme 0.16 mL/g, pH 5.5, and solid-liquid ratio of 1:20, 1.5 h, under the condition of the flavonoids extraction rate can reach 2.74%, the transfer rate of 90.7%.On the pilot scal ,can reach 84.2%. To obtain purification apocynum extract condition of total flavonoids, establish UPLC guide macroporous resin column chromatography method, quickly get optimal elution parameters.Using small gain the key technology parameters, on the pilot scal, the purity was 58.6%, total flavonoids transfer rate was 74%. Explore the method of rapid detection flavonoids content of apocynum leaves , using the prediction model by NIR, get the sample of flavonoids content in 30 seconds, compared with conventional methods for 2 hours, the analysis efficiency can be greatly increased.Stability test results show that the quantitative model to predict the sample standard deviation was 0.10, RSD was 3.62%. Synthesis of quercetin-Cu complexes molecularly imprinted polymer, elutioned by methanol, majority of quercetin may be acquitted, the purity of quercetin calculated according to the area normalization method, the purity of 76.8%.Synthesis of molecularly imprinted polymer rutin, can be used to extract the component structure similar to rutin. |
